Ï€Glutathione S transferase (Ï€GST) enzyme-linked immunosorbent assay

**π-Glutathione S-Transferase (πGST) Enzyme-Linked Immunosorbent Assay (ELISA) Kit** **Purpose:** This ELISA kit is designed for the quantitative determination of π-glutathione S-transferase (πGST) in human serum, plasma, and other biological fluids. It provides a reliable and sensitive method for measuring πGST levels, which can be useful in clinical diagnostics and research applications. **Experimental Principle:** The kit employs a sandwich ELISA technique to detect πGST in the sample. A microtiter plate is pre-coated with a specific monoclonal antibody against πGST. After incubation with the sample, the target πGST binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing, TMB substrate is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow upon addition of an acidic stop solution. The intensity of the color is directly proportional to the concentration of πGST in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the πGST concentration is determined by comparing the OD values to a standard curve. **Kit Composition:** The kit includes all necessary reagents and materials for performing the assay. It is available in both 48-well and 96-well configurations. - **Coated Microplate:** 1 × 48 or 1 × 96 wells - **Standard:** 18 μg/L, 0.5 mL × 1 bottle - **Standard Diluent:** 1.5 mL × 1 bottle - **Enzyme Conjugate:** 3 mL × 1 bottle - **Sample Diluent:** 3 mL × 1 bottle - **TMB Substrate A & B:** 3 mL × 1 bottle each - **Stop Solution:** 3 mL × 1 bottle - **Washing Buffer (20×):** 20 mL × 20 times or 20 mL × 30 times - **Sealing Film:** 2 pieces (for 48-well) or 2 pieces (for 96-well) - **Sealed Bag:** 1 piece **Storage Instructions:** All components should be stored at 2–8°C. The coated plate must be kept sealed and protected from moisture and light. The kit is stable for up to 12 months when stored properly. **Notes:** Ensure all reagents are brought to room temperature before use. Proper pipetting and timing are essential for accurate results. Always include blank and standard wells in the experiment to ensure reliability. This kit is intended for in vitro diagnostic use only.
