This reagent is intended for research use only and is designed to quantify the level of P-selectin (PS) in human serum, plasma, and other related biological samples. The assay is based on the double-antibody sandwich ELISA method. In this procedure, a solid-phase antibody specific to human P-selectin is pre-coated onto the microplate wells. After incubation with the sample, P-selectin present in the specimen binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing to remove unbound components, the substrate TMB (3,3',5,5'-tetramethylbenzidine) is introduced. Under the catalytic action of HRP, TMB turns blue, and upon addition of an acidic stop solution, it changes to a yellow color. The intensity of the yellow color is directly proportional to the concentration of P-selectin in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the P-selectin concentration is determined by comparing the OD values to a standard curve generated from known concentrations.
The kit includes all necessary reagents for performing the assay. It is available in both 48-well and 96-well configurations. Key components include: a coated plate with purified P-selectin antibodies, a set of standards for quantification, a concentrated wash solution, a TMB substrate solution, a developer solution (A and B), and a sample diluent. All reagents should be stored at 2–8°C and are stable under these conditions. Proper storage ensures the integrity and performance of the kit throughout its shelf life.
