**INTENDED USE AND TEST PRINCIPLE**
This FETU-A ELISA kit is specifically designed for research purposes in laboratory settings. It is not intended for use in diagnostic or therapeutic applications. The assay is based on a competitive immunoassay principle. The color change from blue to yellow occurs when the Stop Solution is added, and the intensity of the color is measured at 450 nm using a spectrophotometer. A set of calibration standards is included to generate a standard curve, allowing for accurate quantification of FETU-A levels in the samples.
**SAMPLE COLLECTION AND STORAGE**
- **Serum**: Use a serum separator tube. Allow clotting for 2 hours at room temperature or overnight at 4°C. Centrifuge at 2000×g for 20 minutes. Store aliquots at -20°C. Avoid repeated freeze-thaw cycles.
- **Plasma**: Collect using heparin as an anticoagulant. Centrifuge within 30 minutes at 2000×g, 2–8°C. Store at -20°C.
- **Cell culture supernatants, tissue homogenates, and other biological fluids**: Centrifuge to remove particulates. Assay immediately or store at -20°C. Ensure no hemolysis or granules are present.
**MATERIALS REQUIRED BUT NOT SUPPLIED**
1. 37°C incubator
2. Microplate reader capable of measuring absorbance at 450 nm
3. Precision pipettes, disposable tips, and absorbent paper
4. Distilled or deionized water
**REAGENTS PROVIDED (Storage: 2–8°C)**
- MicroELISA Strip Plate: 12×8 strips / 12×4 strips
- Standard (6 vials): 0.5 ml/vial
- Sample Diluent: 6.0 ml / 3.0 ml
- HRP-Conjugate Reagent: 10.0 ml / 5.0 ml

**NOTES**
- Standard concentrations: 1000, 500, 250, 125, 62.5, 31.2 μg/mL.
- If sample values exceed the highest standard, dilute with Sample Diluent and repeat the test.
- Always use reagents provided by the manufacturer.
- Allow all reagents and samples to reach room temperature (20–25°C) before use. Do not thaw using water baths.
- Never use beyond the expiration date.
- Use only deionized or distilled water for dilutions.
- Keep unused strips in sealed bags with desiccant.
- Use fresh pipette tips for each transfer to avoid cross-contamination.
- All blood-derived products should be treated as potentially infectious. Follow proper biosafety protocols.
- Dispose of samples following local regulations. Allow liquid to stand for 30 minutes to inactivate viruses before disposal.
- Substrate solutions are sensitive; keep away from heat and flame.
- Always read the entire user manual before starting the assay.
**REAGENT PREPARATION**
- Wash Solution (1X): Dilute 1 volume of 20X Wash Solution with 19 volumes of deionized or distilled water. Store at 2–8°C for 1 month.
**ASSAY PROCEDURE**
1. Prepare all reagents and microplate.
2. Add 50 μL of standard or sample to appropriate wells (except blank). Cover with adhesive strip and incubate for 60 minutes at 37°C.
3. Wash the plate 4 times manually or automatically.
4. Add 50 μL of Chromogen A and 50 μL of Chromogen B. Incubate for 15 minutes at 37°C, protecting from light.
5. Add 50 μL of Stop Solution. Read OD at 450 nm.
6. Plot standard curve using average OD values. Determine sample concentration by comparing to the curve.
**QUALITY CONTROL**
- Intra-assay CV < 15%, Inter-assay CV < 15%.
- Assay range: 31.2–1000 μg/mL.
- Sensitivity: <10 μg/mL.
- Cross-reactivity: No significant cross-reaction observed.
**STORAGE**
- Store at 2–8°C for frequent use.
- For long-term storage, keep at -20°C for up to 6 months.
